The biology

The committed step of pyrimidine synthesis, read directly.

Dihydroorotate dehydrogenase oxidises dihydroorotate to orotate, the rate-limiting step of de novo pyrimidine synthesis. Blocking it starves proliferating lymphocytes and tumour cells of nucleotides — the mechanism behind teriflunomide in multiple sclerosis, and behind the next-generation DHODH inhibitors now in Phase 2/3.

The enzyme is usually followed indirectly, through its electron acceptor rather than its product, so anything in a library that reduces the dye reads as activity. Our sensor binds orotate itself, in real time.

Applications

DHODH inhibitor screening

Primary and confirmatory screens on a direct product readout rather than a coupled dye.

Antiproliferative pharmacology

Dose-response and time-course work on pyrimidine-pathway inhibition with a continuous readout.

Redox interference triage

Separate genuine inhibition from redox-cycling compounds that defeat dye-coupled formats.

Other kits

Itaconate Assay Kit

Real-time quantification of a key immunometabolite.

Kynurenic Acid Assay Kit

Direct detection of kynurenic acid without chromatography.
